چکیده
زمینه و هدف: بیماریهای قلبی عروقی در بیماران مبتلا به دیابت یکی از علل اصلی مرگومیر در سراسر جهان هستند. در مقابل، پیشگیری از رتیکولوفاژی قلبی ناشی از دیابت توسط تمرین تناوبی با شدت بالا (HIIT) بهعنوان مداخله غیر دارویی پیشنهادشده است. لذا هدف از پژوهش حاضر بررسی تأثیر HIIT بر محتوای پروتئینهای ATF6،JNK1 و mTOR در بطن چپ قلب موشهای صحرایی دیابتی نوع 2 است.
مواد و روشها: در پژوهش حاضر 12 موش صحرایی نر 2 ماهه از نژاد اسپراگداولی با میانگین وزن 20±180 گرم انتخاب شدند. دیابت نوع 2 از طریق تزریق محلولهای نیکوتینآمید و استرپتوزوتوسین القاء شد. موشهای صحرایی به روش تصادفی به گروه HIIT و کنترل دیابتی تقسیم شدند. برنامه HIIT شامل 5 نوبت 4 دقیقهای با شدت معادل 85-95 درصد حداکثر سرعت و دورههای استراحت فعال 3 دقیقهای با شدت معادل 60-50 درصد حداکثر سرعت بود. برای تجزیهوتحلیل دادهها از آزمون آماری t-مستقل در نرمافزارهای SPSS نسخه 29 استفاده شد. سطح معناداری 05/0P≤ در نظر گرفته شد.
یافتهها: تجزیهوتحلیل دادهها نشان داد که محتوای پروتئین ATF6 و JNK1 پس از 8 هفته HIIT افزایش معنیداری را نسبت به گروه کنترل در بطن چپ قلب نشان داد. درحالیکه محتوای پروتئین mTOR کاهش معنیداری را نشان داد.
نتیجهگیری: به نظر میرسد پروتکل HIIT به دلیل توانایی کاهش استرس شبکه آندوپلاسمی (ER) میتواند برای آزمودنیهای قلبی مبتلابه دیابت نوع 2 توصیه شود.

The Effect of High-Intensity Interval Training (HIIT) on Reticulophagy-Related Proteins in the Left Ventricle of the Heart of Rats with Type 2 Diabetes

Introduction: Cardiovascular diseases in patients with diabetes, particularly through endoplasmic reticulum (ER) stress, are leading causes of mortality worldwide. High-Intensity Interval Training (HIIT) has been proposed as a non-pharmacological intervention to prevent diabetes-induced cardiac apoptosis. This study aims to investigate the effects of HIIT on the expression of ATF6, JNK1, and mTOR proteins in the left ventricle of the hearts of rats with type 2 diabetes.
Materials and Methods: This study was conducted on 12 male Sprague-Dawley rats, aged 2 months, with an average weight of 180 ± 20 grams. Type 2 diabetes was induced through the administration of nicotinamide and streptozotocin. The rats were randomly assigned to either the HIIT group or the diabetic control group. The HIIT program comprised five intervals of 4 minutes at an intensity of 85-95% of maximum speed, interspersed with 3-minute active rest periods at 50-60% of maximum speed. Data was analyzed using independent t-tests in SPSS software version 29, with a significance level of P ≤ 0.05.
Results: Data analysis revealed that the protein content of ATF6 and JNK1 significantly increased after 8 weeks of HIIT compared to the control group, while mTOR protein levels exhibited a significant decrease.
Conclusion: The HIIT protocol appears beneficial for cardiac health in patients with type 2 diabetes due to its capacity to reduce ER stress.
