In this study, the synthesis of nanoparticles of manganese-zinc oxide and zinc iron oxide by chemical co- precipitation method performed and investigation of structural and morphological properties of related nanoparticles using Fourier Infrared spectrum, X-ray diffraction and Transmission electron microscopy were carried out. Antimicrobial effects against shigella flexneri (PTCC 1234) determined by inhibitory diameter zone method. Synergistic effect of nanoparticle with hydrogen peroxide and hypochlorite sodium also assessed by calculation of the fractional inhibitory concentration (FIC) index. Structural analysis of Mn- ZnO and Fe- ZnO nanoparticle revealed that co- precipitation is a suitable method for synthesis of these nanoparticles. Results of antimicrobial tests showed that Mn- ZnO nanoparticles has more antibacterial effect against shigella flexneri than Fe- ZnO nanoparticles. Inhibitory zone diameter of Mn- ZnO and Fe- ZnO were 25 and 14 mm, respectively. In addition, both doped nanoparticle exhibited more antimicrobial effect than zinc oxide nanoparticle so, doping cause increasing antimicrobial effect of zinc oxide nanoparticle. Furthermore, combination of nanoparticles with hydrogen peroxide and hypochlorite sodium showed synergistic effects which was complete synergistic at high concentration and partial synergistic at low concentrations.

The genus Mycobacterium is a group of acid fast bacteria with DNA G+C content of 61-71% and the cell wall containing mycolic acid. According to growth physiology, mycobacteria grouped into two divisions: rapidly-growing and slow-growing mycobacteria. Three groups of mycobacteria exist based on pigment production: nonpigmented (nonphotochromogens), photo-chromogens and scotochromogen. Both human obligate pathogens (M. tuberculosis and M.leprae) and opportunistic species exist in mycobacteria genus. There is different kind of mycobacterial infections such asrespiratory infections, lymphatic node and skeletal involvement, dermal and disseminated infection. Spread of HIV virus and immunodeficiency in recent years makes the mycobacterial infections one of the most dangerous infections. Acid fast staining, biochemical tests and cell wall mycolic acid and fatty acid analysis are used for detection of mycobacteria. Different molecular tests including different PCR basedmethods; hybridization and sequencing tests are used for diagnosis and also verifying phenotypic and biochemical tests. Clarithromycin, rifabutin, ciprofloxacin, rifampin and ethambutol currently were used for treatment of slow growing mycobacterial infections. Whereas macrolides, cephalosporins and fluoroquinolones are used for rapidly growing treatment. For rapidly growing mycobacteria, disk diffusion method, disk elution method and broth micro dilution method are more appropriate. Radiometric methods and broth micro dilution method are choice methods for slow growing mycobacteria drug susceptibility test. Moreover molecular methods like PCR single-strand conformational polymorphism (PCR-SSCP), Sequencing, PCR-RFLP and Multiplex-PCR are developed for drug susceptibility determination in mycobacteria. According to non-tuberculosis mycobacteria clinical significance, clinical laboratories and health care team most take attention to diagnosis of these bacterial infections.

Abstract:Aims: Collagen synthesis can help to heal wounds, especially Large wounds. Any treatments that can stimulate collagen synthesis in fibroblasts can accelerate wound repair, so it has clinical value. Stem cells secrete some macromolecules and signals that can have different effects in other cells.Materials and Methods: In this study, human mesenchymal stem cells were cultured and their medium supernatant was harvested as a medium containing the signal. After determining concentration, they were exposed to mouse fibroblasts. The cell viability was examined and then in the molecular section, by designing specific primers, three The collagen gene was evaluated by Real-Time PCR.Results: The results indicated a growth-promoting effect using the amount of 20 μg /ml of stem cells conditioned media in the culture medium of fibroblast cells. It was also found in the molecular section, that this concentration could increase the expression of all three selected collagen genes.Discussion and conclusion: The results of this study fully confirmed the initial goals, indicating the high power of mesenchymal stem cells in the induction of collagen synthesis, and thus could be a very good candidate for clinical studies.
