Abstract
Background & Objectives: Metastatic cancers, such as breast cancer, are generally resistant to chemotherapy. Therefore many researches try to find new and effective compounds to treat this type of cancer. In this study, we examined the Silibinin toxic effect on tumor growth and proliferation of MCF-7 cell line.
Materials & methods MTT assay was used to assess the inhibitory effect of silibinin on metabolic activity of MCF_7 cells. For this purpose, seeded cells were plated on to 96-well plates and then exposed to varying concentrations of silibinin (0-350 ug/mL) for different time (24, 48 and 72 h). The percentage of metabolic activity was calculated according to the MTT test. In next step, in order to evaluate the effect of silibinin on apoptosis in MCF_7 cells, Caspase 3/7 test was used.
Results: Our results indicate that silibinin inhibited metabolic activity of MCF-7cells in a dose-dependent manner and it can induce apoptosis pathway in these cells.
Conclusion: The study showed that treatment of MCF-7 cell line with Silibinin can be through induction of apoptosis, inhabitation of the growth and proliferation of cells. Due to the significant inhibitory effect of silibinin on MCF-7 cells, it seems important for researchers to utilize milk thistle in the treatment and prevention of cancer.

چکیده
زمینه و هدف:سرطانهای متاستاتیک مثل سرطان پستان، عموما نسبت به شیمیدرمانی مقاوم هستند؛ لذا یافتن ترکیبات مؤثر و جدید برای درمان این نوع سرطانها مورد توجه محققین قرار دارد. در این مطالعه اثر سمی سیلیبینین بر رشد و تکثیر رده سرطانی MCF-7 بررسی شده است.
مواد و روشها: بهمنظور بررسی اثر مهاری سیلیبینین بر فعالیت متابولیکی سلولهای MCF-7 تست MTT انجام شد. برای این منظور سلولها در پلیت 96 خانه کشت داده شدند و سپس در معرض غلظتهای مختلف سیلیبینین (350-0) میکروگرم بر میلیلیتر در زمانهای متفاوت 72-48-24 ساعت قرار گرفتند و به این ترتیب درصد فعالیت متابولیکی سلول ها محاسبه شد، همچنین بهمنظور بررسی تأثیر سیلیبینین بر آپوپتوز تست کاسپاز 3 و 7 انجام شد.
یافتهها:نتایج این تحقیق نشان داد سیلیبینین باعث مهار فعالیت متابولیکی رده سلولی MCF-7 بهصورت وابسته به دوز و زمان میشود، همچنین آپوپتوز را در این سلولها القا میکند.
نتیجهگیری: این تحقیق نشان داد که تیمار رده سلولی MCF-7 با سیلیبینین میتواند از طریق القای آپوپتوز، سبب مهار رشد و تکثیر شود. با توجه به تأثیر مهاری معناداری که تیمار سلولها با سیلیبینین بهدنبال دارد، بهنظر میرسد که زمینه تحقیقاتی مناسبی برای بهرهبرداری از گیاه خار مریم در کنترل و درمان سرطانها وجود دارد.

مقدمه
سرطان پستان یکی از شایعترین بیماریها در سراسر جهان و بهعنوان دومین علت مرگومیر ناشی از سرطان در زنان شناخته میشود (2,1). شیوع این سرطان به حدی است که گفته میشود در برخی کشورهای غربی از هر 7 نفر خانم یک نفر در طول عمر خود دچار سرطان پستان خواهد شد این مهم بهویژه در بانوان میانسال نگران کنندهتر است (4,3).
از عوامل اساسی بروز سرطان میتوان به عوامل محیطی از جمله آلودگی هوا، استرس، الگوی زندگی و رژیم غذایی افراد اشاره کرد که با ایجاد جهش و تغییرات ژنتیکی، مسـئول بروز بدخیمیها هستند (7-5). از طرفی دیگـر، علیرغم استفاده از راهکارهای درمانی از جمله جراحی، شـیمیدرمانی و رادیوتراپی همچنان میزان مرگومیر در بیماران مبتلا بالا میباشد که خود حکایت از ناکارآمدی این راهکارهای درمانی دارد. به علاوه، تأثیر مخرب شیمیدرمانی و پرتودرمانی بر سلولهای نرمال در حال تقسیم نیز از جمله معایب دیگر ایـن نوع درمانها محسوب میشود. مشخص شده است که مصرف مواد غذایی که دارای خاصیت آنتیاکسیدانی است، در پیشگیری و کاهش ابتلاء به سرطانها نقش مؤثری دارد. لذا با توجه به موارد اشاره شده، تمایل به استفاده از فراوردههای گیاهی دارای خاصیت آنتیاکسیدانی طی سالهای اخیر بسیار افزایش داشته است. در این میان آنتیاکسیدانهای پلیفنول طبیعی، بهعلت کارایی بالا و تأثیر جانبی کم بهعنوان یکی از مؤثرترین ترکیبات ضدسرطان شناسایی شدهاند (9,8).
سیلیمارین یک فلاوونوئیـد پلیفنولیک است که از دانههای گیاه خارمریم (milk thistle) استخراج شده و حدود 90 درصد از آن را ترکیبی به نام سیلیبینین (silibinin) به خود اختصاص میدهد. سیلیبینین ناهماهنگی بین بقای سلول و آپوپتوز را از طریق دخالت در بیان ژنهای دخیل در چرخهی سلولی و آپوپتوز، تنظیم میکند. بررسیهای مهار رشد و تکثیر سلولهای سرطانی روی سیلیبینین مؤید نقش آن بهعنوان ترکیبی ضد سرطانی میباشد (12-10). نقش ضد سرطانی این ترکیب روی ریه (13)، تخمدان (14)، گلیوبلاسـتوما (15)، پروستات (16)، مثانه (17)، کبد (18)، پستان (19) و کلون (20) گزارش شده است.
مواد و روشها
کشت سلول
رده سلولی MCF-7 از بانک سلولی انسیتو پاستور (تهران، ایران) تهیه شد. سلولها در محیط کشت RPMI1640 (Gibco) همراه با یک درصد آنتی بیوتیک پنیسیلین- استرپتومایسین (Sigma) و ده درصد سرم جنین گاوی (Gibco) کشت داده شدند و در انکوباتور °C 37 و فشار %5 از CO2 رشد داده شدند.
تیمار سلولها و سنجش تکثیرسلولی
ابتدا سلولهای MCF-7 ، با غلظتهای 50 ، 75 ، 100 ، 150 ، 200 ، 250 ، 300 و 350 میکروگرم بر میلیلیتر از داروی سیلیبینین تیمار شدند، سپس بهترتیب پس از زمانهای 24 ، 48 و 72 ساعت مورد مطالعه قرار گرفتند.
جهت اندازهگیری توانحیاتی سلول و اثرات سمی سیلیبینین بر رشد و تکثیر رده سلولی MCF-7، از تست MTT استفاده شد. در اجرای این روش، بهترتیب زیر عمل شده است: ابتدا میزان 7000 سلول در هر یک از چاهکهای پلیت 96 خانه کاشته شد و بهمدت 24 ساعت به منظور چسبیدن به کف پلیت و رسیدن به تراکم 70 درصد به آنها زمان داده شد، سپس با غلظتهای مذکور از سیلیبینین تیمار و پس از زمانهای 24 ، 48 و 72 ساعت، محیط هر چاهک با 100 میکرولیتر محلول متیل تیازولتترازولیوم (MTT) تعویض و بهمدت 4 ساعت انکوبه شد. سپس بلورهای فورمازان حاصل در 100 میکرولیتر دی متیل سولفوکساید حل وجذب آن توسط دستگاه ELISA-reader )مدل power wave xs2 ساخت شرکت BioTek (، در طول موج 570 نانومتر اندازهگیری شد (با سه بار تکرار). برای محاسبه اثر توکسیسیتی داروی سلیبینین بر روی بقاء سلولهای MCF-7 و بررسی میزان مرگ سلولهای تیمار شده از فرمول زیر استفاده شد:
100 × = درصد توانایی حیاتی سلولها
در این فرمول، OD exp و OD cont بهترتیب بیانگر جذب نوری سلولهای تیمار شده و سلولهای تیمار نشده (کنترل) میباشد. پس از اندازهگیری جذب نوری محلولها IC50 سلولها در هر بازه ی زمانی محاسبه شد.
سنجش فعالیت کاسپاز3 و7
در این مرحله از کیت کاسپاز 3 و 7 شرکت Promega استفاده شده است. این کیت میزان لومینسانس ایجاد شده در اثر فعالیت آنزیمهای کاسپاز ۳ و ۷ را اندازهگیری میکند. کاسپازها از پروتئازهای سیستئین- آسپارتیک اسید هستند و نقشی اساسی در آپوپتوز سلولهای پستانداران دارند. کیت کاسپاز ۳ و ۷ حاوی پروتئین آمینولوسیفرین متصل شده به تتراپپتید DEVD (آسپارتیک اسید، گلوتامین، والین، آسپارتیک اسید) میباشد که بهعنوان پیش ماده اختصاصی برای کاسپازهای ۳ و ۷ عمل میکند. همچنین، این کیت دارای بافری مناسب برای فعالیت کاسپاز و لوسیفراز است. افزودن این بافر به سلولها موجب تجزیه آنها و سپس جداشدن تتراپپتید از آمینولوسیفرین، در اثر فعالیت کاسپازها میشود. بهدنبال آن، آمینولوسیفرین بهعنوان پیش ماده لوسیفراز عمل کرده و این آنزیم باعث تجزیه آن شده و لومینسانس تولید میکند. پرتونوری تولیدشده متناسب با میزان فعالیت کاسپازهای تولیدشده در سلولهای در حال آپوپتوز بوده و توسط دستگاه لومینومتر در دامنه ٤٠٠ تا ٧٠٠ نانومتر آشکارسازی میشود. برای این منظور سلولها را در فلاسک T-25 کاشته و پس از 24 ساعت که تراکم آنها به 70 درصد رسید، بر اساس نتایج بهدست آمده از تست MTT ، سلولها را با دوزهای 75 ، 100 ، 150 و 200 میکروگرم بر میلیلیتر از سیلیبینین تیمار کرده و سپس به مدت 48 ساعت درون انکوباتور قرار داده شدند. پس از طی این زمان با اضافه کردن بافرلیز سلولی به هر فلاسک سلولها بهصورت سوسپانسیون در یـخ، بهمدت ده دقیقـه انکوبـه و سانتریفوژ (1 دقیقــه و ×g 10000) گردیدند. سپس 5 میکرولیتر از معرف با سوپرناتانت حاصل از سانتریفوژ، ادغام شده و و پلیت در انکوبــاتور 37 درجه سانتیگراد بهمدت دو ساعت انکوبه شد. شکست کروموفور توسط کاسپاز 3 و 7 ، مؤید فعالیت آنزیم میباشد که با استفاده از دستگاه لومینومتر میزان نشر نور اندازهگیری شد. برای دقت بیشتر نتایج، هر تیمار سلولی، سه بار کشت داده شده و میزان جذب آن خوانده شده است.
از نرمافزار Graphpad (prism 6) برای آنالیز دادههای حاصل از MTT و تست کاسپاز 3 و7 در دوزها و زمانهای مختلف استفاده شد و معناداری نتایج مورد بررسی قرار گرفت. مقادیر بهدست آمده با 05/0p< از نظر آماری معنادار بود.
یافتهها
نتایج تأثیر سیلیبینین بر توانحیاتی سلولها حاکی از آن بود که غلظتهای 50 ، 75 ، 100 ، 150 ، 200 ، 250 ، 300 و 350 میکروگرم بر میلیلیتر نسبت به گروه کنترل سبب مهار رشد سلولها شدند. در غلظتهای پایین سیلیبینین (50 و 75 میکروگرم بر میلیلیتر) کاهش معناداری در توان زیستی سلولها مشاهده نشد. مقایسه دادهها دردوزهای مورد استفاده برای سیلیبینین برای زمانهای مختلف نشان داد که روند کاهش درصد توان زیستی سلولها با افزایش غلظت و با گذشت زمان همراه است (شکل 1).
همچنین برای هر یک از زمانهای 24، 48، 72 ساعت دوزی از سیلیبینین که بهواسطه آن 50 درصد سلولها زنده میمانند محاسبه و تعیین شد (جدول 1).
شکست پروتئولیتیک پروتئینهای سلولی بـا ارزیابی فعالیت کاسپاز مشخص شد، بهطوریکه در بالاترین غلظت سیلیبینین استفاده شده در این مطالعه (200 میکروگرم بر میلیلیتر) طی مدت زمان 48 ساعت پس از تیمار ، بیشترین فعالیت کاسـپاز 3و 7 مشـخص گردیـد. پـس بـا افـزایش غلظت سیلیبینین افزایش معناداری در فعالیت کاسپاز ایجاد شد که مؤید افزایش فعالیت مولکولهای پروآپوپتوزی در برابر محرک های القایی آپوپتـوز (سیلیبینین) میباشد (شکل 3).
بحث
در این پروژه نشان داده شد که تیمار رده سرطانی MCF-7 با سیلیبینین میتواند سبب مهار رشد سلولهای MCF-7 شود. در مطالعاتی که در سال 2014 توسط یوسفی و همکاران صورت گرفت نشان داده شد که سیلیبینین سبب مهار تکثیر سلولهای سرطانی SKBR3 میگردد و با افزایش غلظت بهکار رفته اثر مهار آن نیز افزایش مییابد (21). در همین سال توسط Reyhan Akhtar و همکاران نشان داده شد که سیلیبینین دارای خواص ضدسرطانی بر روی ردهی سرطانی کولون HT-29 می باشد. همچنین سیلیبینین بهطور مؤثری سبب مهار رشد سلولهای سرطانی پروستات ردههای 1AsPC- ، 3BxPC- و 1Panc- میشود که با القاء آپپتوز همراه است (22). همراستا با نتایج حاصل از تحقیقات سایرین، نتـایج حاصـل از این پروژه نشان میدهد که سیلیبینین بر روی رده سرطانی MCF-7 اثر سمی وابسته به دوز و زمان دارد. با توجه به نتایج MTT سیلیبین در غلظتهای بیشتر از 75 میکروگرم بر میلیلیتر پس از 48 ساعت براین رده سلولی اثر مهارکنندگی بر تکثیر سلولی دارد. همچنین مقایسه نتایج حاصل از تست MTT و تست کاسپاز نشان داد که در غلظتهای بالای سیلیبینین (بیش از 75 میکروگرم بر میلیلیتر) که میزان فعالیت کاسپاز در آنها بالاتر میباشد، باعث کاهش معناداری در میزان زنده ماندن سلولها در نتایج حاصل از تست MTT نیز میشوند. بهنظر میرسد القای مرگ برنامهریزی شده سلولی از مهمترین مکانیسمهای مؤثر در فعالیت ضدسرطانی سیلیبینین میباشد که یافتههای این بررسی آن را تأیید میکند .
در مجموع، نتایج حاصل از پژوهش انجام شده مؤید خاصیت پروآپوپتوتیک سیلیبینین و اثربخشی این ترکیب گیاهی علیه رده سرطانی MCF-7 میباشد. با توجه به اینکه نتایج این تحقیق صرفاً روی رده سرطانی MCF-7 انجام شده است، تأثیر این ترکیب روی سلولهای طبیعی نیازمند بررسی بیشتر میباشد.
