چکیده

زمینه و هدف  سرطان تخمدان، یکی از کشنده‌ترین بدخیمی‌های زنان است؛ بنابراین، ضرورت کشف روش‌های تشخیصی و درمانی جدید احساس می‌شود. استفاده از بیومارکرهای جدید برای تشخیص سریع‌تر و انتخاب ملاحظات درمانی بهتر، یکی از دغدغه‌های اصلی در این زمینه است. مطالعات اندکی در مورد عملکرد انکوژن بودن یا مهارکنندگی تومور miR-4443 در سرطان انجام شده است. بنابراین برای مشخص شدن نقش این miRNAs در سرطان تخمدان، هدف مطالعه حاضر، بررسی میزان بیان miR-4443 در نمونه‌های بافتی سرطان تخمدان و ارتباط آنها با فاکتورهای بالینی بوده است.
مواد و روش‌ها در مطالعه حاضر که یک مطالعه توصیفی- تحلیلی است، با توجه به نقش miR-4443، سطح بیان آن توسط روش qPCR در 35 نمونه توموری تخمدان و 35 نمونه غیرتوموری پارافینه، بررسی شد. بررسی بیان توسط پرایمرهای اختصاصی miRNA انجام شد و سپس آنالیز آماری برای مشخص شدن معناداری صورت گرفت. در مرحله بعدی، ارتباط فاکتورهای بالینی با میزان بیانmiRNAs سنجیده شد.
یافته‌ها نتایج حاصل از مطالعه نشان داد که بیان miR-4443 در نمونه‌های توموری در مقایسه با نمونه‌های غیرتوموری، کاهش می‌یابد (P<0.0001) و این کاهش بیان با متاستاز نیز مشاهده شد.
نتیجه‌گیری با توجه به نتایج، مطالعه حاضر نشان داد که miR-4443 نقش مهارکنندگی تومور را دارد، احتمالاً کاهش در بیان آن با افزایش تکثیر و تهاجم سلولی همراه است و با بررسی بیشتر، علاوه بر مطالعه حاضر می‌توان آن را به عنوان بیومارکرهای تشخیصی یا هدف‌های درمانی در سرطان تخمدان پیشنهاد داد.

Study of the expression level of microRNA-4443 in tissue samples paraffinembedded ovarian cancer comparison with non-tumor samples

Introduction: Ovarian cancer (OC) is one of the most lethal malignancies in women. Hence, new investigative and therapeutic methods for OC are instantly needed. The use of new biomarkers for faster diagnosis and selection of more efficient therapies is one of the main concerns in this area. Few studies have been conducted on the effects of miR-4443 in cancer. Therefore, to determine the role of thismiRNA in OC, this study was directed to investigate the expression rate in OC tissue samples and its relationship with clinical factors.
Materials and Methods: In this study that was descriptive-analytic, regarding the role of miR-4443, its expression levels were evaluated in 35 ovarian tumor and 35 ovarian non-tumor tissue samples paraffinembedded by using qPCR. Expression was investigated by miRNA specific primers and then statistical analysis was performed to determine the significance. In the next step, the relationship between clinopathologic factors and miRNA expression was investigated. Results: The results of the study showed that miR-4443 decreased in OC and in metastatic samples (P<0.0001). Conclusion: As a result, this studied miRNA may contribute to suppressing tumor. Therefore decrease in its expression is associated with increased cell proliferation and invasion. Further investigation can help to suggest thismiRNA as diagnostic biomarker or therapeutic target in OC.
