چکیده

زمینه و هدف: فاکتور VII یکی از عوامل انعقادی مهم در مسیر خارجی انعقاد خون است که می تواند با فعال کردن FX نیاز بیماران هموفیل را به فاکتورهای VIII و IX رفع نماید. بیان نو ترکیب این فاکتور مشکلات موجود در تهیه فاکتور های مذکور از پلاسما (با توجه به میزان اندک آن ها) و همچنین خطر انتقال بیماری های خونی را برطرف می سازد. لذا هدف از این پژوهش بیان فاکتور مذکور به صورت نو ترکیب و در سطح بالا با استفاده از فناوری Gateway و TOPO cloning می باشد.
مواد و روش ها: در این تحقیق تجربی، cDNA فاکتور VII از رده سلولی کبدی HepG2 به کمک PCR جدا شد و سپس با تکنیک TOPO Cloning ژن مورد نظر در ساختمان ناقل پروکاریوتی قرارگرفت. ناقل نوترکیب پس از غربالگری برای کلونی های باکتریایی استخراج و از آن برای انجام واکنش نوترکیبی LR با ناقل بیانی باکولوویروس در فناوری Gateway استفاده شد. ویروس نوترکیب به میزبان حشره انتقال و پس از غربالگری های لازم بیان پروتئین بررسی گردید. نتایج آنالیز بیان پروتئین با الایزا به صورت تکرار سه تایی (انحراف معیار ± میانگین) ارایه و تفاوت بین گروه ها با استفاده از آزمون آماری تی استیودنت تحت نرم افزار SPSS مورد مطالعه قرار گرفت.
یافته ها: نتایج حاصل از تست PCR برای واکنش های کلونینگ و نو ترکیبی همگی حاکی از کلونینگ ژن فاکتور VII با بازدهی بیش از 90 درصد در ساختار ناقل های مورد استفاده بودند. آنالیز های انجام شده برای بیان پروتئین توسط الایزا، SDS-PAGE و وسترن بلات بیان بالای این فاکتور را (30 mg/mL) تایید نمودند. نتایج حاصل از آنالیز الایزای نمونه های کشت آلوده تفاوت معناداری را با کنترل منفی نشان می دادند (P

Background and purpose: Factor VII is one of the important coagulation factors in extrinsic blood coagulation pathway which can resolve the use of FVIII and FIX for hemophilia patients by activating FX. Recombinant expression of this factor can eliminate the potential problems in preparing those factors from plasma and the risk of transferring hematological diseases. Therefore the present study intended to investigate the expression of recombinant FVII at a higher level using Gateway technology and TOPO cloning. Methods and Materials: In this experimental study Factor VII cDNA was isolated from HepG2 cell line by PCR and cloned to prokaryote TOPO vector by TOPO cloning reaction. The recombinant vector was extracted for bacterial colonies after screening and was used in Gateway adapted Baculovirus DNA by LR recombination reaction. The recombinant virus was transfected onto insect cell line and the expression of the protein was analyzed after necessary screening. Findings of the protein expression via ELISA were presented in triadic (Mean ± SD); the differences across the three groups were investigated using Student t-test. Results: Cloning and recombination reaction analysis by PCR determined cloning of rFVII in high accuracy (≥90%) in the vectors. High level expression of recombinant FVII was confirmed by SDS-PAGE ELISA and Western blot analysis (30μg/ml).
