چکیده

زمینه و هدف: ویتامین ها با کاهش رادیکال های آزاد نقش موثری در بهبود بیماری های عصبی دارند، لذا در این تحقیق اثر ترکیبی والپروات سدیم با ویتامین C و E بر تشنج در موش صحرایی بررسی شد.
مواد و روش: 49 سر موش صحرایی به 7 گروه تقسیم شدند: گروه کنترل (سالین: 1 میلی لیتر بر کیلوگرم)، گروه والپروات سدیم (100 یا 200 میلی گرم بر کیلوگرم)، گروه ویتامین C (250 میلی گرم بر کیلوگرم)، گروه ویتامین E 100( میلی گرم بر کیلوگرم) و گروه ویتامین C یا ویتامین E با والپروات سدیم. استریکنین به صورت زیر پوستی 30 دقیقه بعد از دریافت درون صفاقی داروها یا سالین، تزریق شد. سپس مدت زمان شروع تشنج، مدت زمان مرگ و میزان زنده ماندن طی 30 دقیقه ثبت گردید.
یافته ها: والپروات سدیم با دوز 100 و 200 میلی گرم بر کیلو گرم زمان شروع تشنج و مدت زمان تا مرگ را نسبت به گروه کنترل به صورت معنی داری به تاخیر انداخت و میزان زنده ماندن به ترتیب 36% و 100% بود.کاربرد توام والپروات سدیم و ویتامین C زمان شروع تشنج و مدت زمان مرگ را به طور معنی داری نسبت به گروه کنترل افزایش داد و 57% موش ها زنده ماند. کاربرد توام ویتامین E با و والپروات سدیم زمان شروع تشنج و مدت زمان مرگ را به طور معنی داری نسبت به گروه کنترل به تاخیر انداخت و 66% موش ها زنده ماندند.
نتیجه گیری: کاربرد ویتامین C و E با والپروات سدیم تشنج القا شده توسط استریکنین در موش های صحرایی را کاهش داد.

Therapeutic effects of combination of sodium valproate and vitamin C and E on seizure induced by strychnine in adult rat

Background: Vitamins play an important role in the improvement of neurological diseases by reducing free radicals, therefore, in this study, the effect of combine therapy sodium valproate with vitamin C and E was evaluated on seizure in adult rat.
Material and methods: 49 rats were divided to 7 groups consisting of control group (saline: 1ml/kg (, sodium valproate groups (100 or 200 mg/kg sodium valproate), Vitamin C group (250 mg/kg), vitamin E group (100 mg/kg) and vitamin C or vitamin E with sodium valproate groups. The animals received a subcutaneous injection of strychnine for induction of convulsive seizures, 30 minutes after intraperitoneal administration of drugs or saline. Then convulsion onset time and death time and survival rate were recorded.
Results: Sodium valproate with doses of 100 and 200 mg/kg delayed the convulsion onset time and death time significantly compared to the control group and the survival rate was 36% and 100% respectively. The combination of sodium valproate and vitamin C significantly increased the time to start seizure and death time than the control group. Also the survival rate was 57%. The combination of sodium valproate and vitamin E increased the onset of seizure and the duration of death compared to the control group and 66% of the rats survived.
Conclusion: The use of vitamin C and E with sodium valproate has a significant role in reducing seizure induced by strychnine in adult rat.
