بررسی بیان دو ژن از خانواده ی هموباکس در سرطان پستان

چکیده

چکیده:
مقدمه : سرطان پستان یکی از کشنده ترین سرطان ها در بین زنان میباشد . این نوع سرطان یک بیماری به شدت ناهمگن است وشامل LCIS ،(Lobular carcinoma in situ) ،(Ductal carcinoma in situ) DCIS و کارسینوم مهاجم میشود. شناسایی راه های مولکولی می تواند در پیشگیری و راه های درمان کمک کننده باشد در این مطالعه بمنظور مسیر Development در این سرطان، بیان دو ژن CDX1 ,2بررسی گردید .
روش کار :در این مطالعه در ابتدا استخراج RNA توسط کیت های استخراج توتال RNA از40 بافت تومری و 40 بافت نرمال در همان بیمار صورت گرفته و سپس سنتز cDNA توسط کیت تاکارا صورت گرفته و در ادامه میزان بیان ژن CDX1 , CDX2 توسط تکنیک Real time PCR ارزیابی شد .
نتایج :نتایج ما نشان داد که بیان ژن CDX1در بافت های توموری مبتلایان به سرطان پستان به میزان 50 درصد و CDX2 به میزان 45 درصد بصورت معنی دار کاهش پیدا کرده است. همچنین مطالعات آماری ما نشان داد که کاهش بیان CDX1 با سایز تومور ارتباط مستقمی دارد.
بحث و نتیجه گیری :مسیرهای مختلف سیگنالینگ سرطان ها را تحت تاثیر قرار میدهند، با توجه به نتایج ما در این مطالعه، کاهش بیان دو ژن CDX1,2 در این سرطان نشان دهنده نقش تومور ساپرسوری آن ها می
باشد. مطالعات بیشتری نیاز می باشد تا نقش دقیق این دو ژن را در سرطان پستان روشن نماید.

Introduction: Breast cancer is one of the main cause of death in women in the world. This cancer is heterogeneous and there are three types including LCIS ،(Lobular carcinoma in situ) ،(Ductal carcinoma in situ) DCIS and carcinoma. Although there are chemotherapy and surgery for its treatment, the molecular mechanism help to diagnosis in the first stage. In this study we aimed to show the expression of CDX1 and CDX2 in breast cancer.
Materials and methods:
In this study, total RNA was extracted from 40 tumor and 40 normal tissues using RNA extraction kit. After cDNA synthesis with Takara cDNA synthesis kit, expressional analysis of CDX1 and CDX2 gene was evaluated by Real time PCR techniques.
Results:
Our data showed the expression of CDX1 and CDX2 gene were decreased in this cancer, 50 and 45 percent, respectively. Also our statistical analysis demonstrated the underexpression of CDX1 is correlated with tumor size.
Discussion: To the best our knowledge, this is the first study that shows the expression of these gene were reduced in breast cancer. We suggest that these genes have tumor suppressor role in breast cancer and it require more studies to show the main mechanism of these genes in breast cancer
